What a fantastic question!
This is tough one.
I'm totally torn, but a little part of me leans towards Velma- because the all-fantastic BeBe played this part and I got to see her. She was spectacular.

I too am having difficulty choosing..they are both amazing..probably Velma..because she is so sassy..and I'd totally Kill to play her (literally..jk)

Velma is way cooler, Roxie actually kinda bugs me as a character...the kinda person I'd hate with a vengeance if I ever came in contact with.

Velma because she's just an all-over more interesting character. She's a conniving wench and she knows it. Plus, she gets a ton of great songs.

Velma.
The role of Velma requires acting skills, the wit and humor of the role is not in the words all the time, the actress playing the role has to find the humor and make it funny. With Roxie, the humor is mostly there in the script already. Even a not-so-good-actress (Denise Van Outen for example!!! She was awful!!) can pull off Roxie, but not Velma.
